Dr. Lee's expertise are in the evolution of mitochondrial genetic systems in green algae. The mitochondrial DNAs (mtDNA) of Chlamydomonas algae contrast with those of land plants and the few other green algal mtDNAs that have been characterized in detail. The unusual Chlamydomonas features include discontinuous and scrambled mitochondrial rRNA coding regions which encode fragmented mitochondrial rRNA, reduced mitochondrial gene content and a more rapid rate of nucleotide substitution among mitochondrial genes. By examining these features, particularly those of discontinuous mitochondrial rRNA gene structure and fragmented mitochondrial rRNA, among the class of Chlamydomonas algae and other related green algal taxa, Dr. Lee's research group hopes to determine the phylogenetic stage(s) when these differences arose and ultimately the factors that brought these changes about.

Other interests include phylogenetic analysis of mitochondrial gene sequences, characterization of green algal mitochondrial ribosomes and recombination of mtDNA.
